ABSTRACT

Methanol extract of Moringa oleifera leaves can function as an antioxidant, antiproliferation, or antitumor. This study aims to prove the effect of moringa leaf methanol extract, especially NTT varieties on the expression of apoptotic proteins (TRAIL R1, Bcl-2, and caspase-3) in DMBA-induced wistar rat colon tissue. The experimental design of post test group design was carried out on 30 wistar rats which were selected at random randomly. All rats were conditioned in a homogeneous state and divided into 5 treatment groups based on the dose of methanol extract of kelor var leaves of NTT and DMBA which were given orally and then observed for 60 days. Immunohistochemical TRAIL-R1 and Bcl-2 examination (Anti human / mouse / rat Bcl-2 monoclonal IgG2 purified mouse and monoclonal IgG2A Anti-m TRAIL R1 purified Rat). Caspase-3 examination with the ELISA (human / mouse caspase-3 Duoset IC® ELISA R & D System) method. Data were analyzed by ANOVA test and correlation. The results showed the administration of methanol extract of Moringa var leaves NTT gave a significantly lower TRAIL R1 mean at doses of 20 and 40 mg / KgBB but not at a dose of 80 mg / KgBB. The three doses also gave lower mean Bcl-2 than positive controls. There was no significant mean caspase-3 relationship between treatments with (F = 0.375, p = 0.824). It can be concluded that the administration of methanol extract of Moringa leaves of NTT variants can reduce the expression of TRAIL-R1 at doses of 20 and 40mg / KgBB and expression of Bcl-2 at all doses but did not affect the mean levels of caspase-3 in DMBA-induced rat colon tissue.
